硫代葡萄糖苷

硫代葡萄糖苷是许多辛辣植物的天然成分,如芥末卷心菜辣根。 这些植物的刺激性是由于当植物材料被咀嚼,切割或以其他方式损坏时由芥子油苷产生的芥子油。 这些天然化学物质最有可能有助于植物防御病虫害,并赋予十字花科蔬菜特有的苦味。[1]

硫代葡萄糖苷是几乎所有芸苔属植物的次生代谢产物,例如芸苔目中有重要经济用途的十字花科山柑科番木瓜科。 在芸苔属植物之外,非洲核果木科中的Drypetes属[2]和Putranjiva属是唯一已知含有硫代葡萄糖苷的属。 硫代葡萄糖苷存在于各种可食用植物中,例如卷心菜类(白卷心菜,大白菜西兰花),西洋菜辣根刺山柑萝卜,其中分解产物通常贡献了它们的独特味道。 硫代葡萄糖苷也存在于这些植物的种子中。[1]

硫代葡萄糖苷衍生自葡萄糖氨基酸,构成一类含有的天然有机化合物。它们是水溶性阴离子,属于葡萄糖苷。每个硫代葡萄糖苷含有中心碳原子,其通过硫原子与硫代葡萄糖基团结合,并通过氮原子与硫酸基团结合(形成硫酸化醛肟)。另外,中心碳与侧基结合;不同的硫代葡萄糖苷具有不同的侧基,并且侧基的变化是造成这些植物化合物的生物活性变化的原因。硫代葡萄糖苷的半系统命名由该侧链的化学名称和“硫代葡萄糖苷”组成,例如烯丙基芥子油苷与烯丙基硫代葡萄糖苷。[3] 硫代葡萄糖苷化学的实质是硫代葡萄糖苷在黑芥子酶酶水解硫代葡萄糖苷键后转化为异硫氰酸酯(“芥子油”)的能力。[3]

  • (R)-2-羟基丁-3-烯基芥子油苷(progoitrin)可能是(S)-2-羟基丁-3-烯基异硫氰酸酯的前体,预计它是不稳定的,并会立即环化形成(S)-5-乙烯基𫫇唑烷-2-硫酮(甲壳素)

已知大约132种不同的硫代葡萄糖苷在植物中天然存在。 它们由某些氨基酸合成,如脂肪族硫代葡萄糖苷,主要来源于蛋氨酸,其余还有丙氨酸亮氨酸异亮氨酸缬氨酸。 (大多数硫代葡萄糖苷实际上衍生自这些氨基酸的链延长同源物,例如,萝卜硫苷衍生自二高蛋氨酸,其是甲硫氨酸链延长两次)。芳香硫代葡萄糖苷包括衍生自色氨酸的吲哚,例如来自色氨酸的芥子油苷,还有的来自苯丙氨酸,其链延长的同系物高苯丙氨酸,和来自酪氨酸的sinalbin。[4]

在有水的条件下,植物中的黑芥子酶会从硫代葡萄糖苷中切除葡萄糖基团。[5] 然后剩余的分子迅速转化为异硫氰酸酯硫氰酸盐;这些是植物自我防御的活性物质。 硫代葡萄糖苷也称为芥子油糖苷,由于此反应的标准产物是异硫氰酸酯(芥子油); 另外两种产物主要于含有专门改变反应结果的植物蛋白质的条件下产生。[6]

在上文说明的化学反应中,与实际相比,图左侧的红色弯曲箭头被简化了,因为未显示酶黑芥子酶的作用。 然而,所示的机理基本上与酶催化反应一致。

益处 编辑

食用较多十字花科蔬菜的人群似乎癌症发病率、老年痴呆率都较低,硫苷及异氰酸盐分解产物可能为有效成分之一。[7][8][9]

硫苷及分解产物在动物和细胞模型中有抗肿瘤、抗氧化、抗痴呆等作用,但直接加入人类饮食中的益处尚不明确。[10]

毒性 编辑

如果硫代葡萄糖苷的浓度高于一种动物可接受的浓度,那么含硫代葡萄糖苷的作物作为动物的主要食物来源会产生负面影响。硫苷的分解产物中,异氰酸盐造成苦味降低摄食,腈基物质可能影响健康,[11]硫氰酸及其酯已被证实有抑制碘吸收的抗甲状腺素作用,会造成甲状腺肿大[12] 然而,即使在同一属(例如开罗刺鼠金刺鼠)中,对硫代葡萄糖苷的植物耐受水平也不同。[13]

菜籽油压榨剩下的油渣饼富集了菜籽中大部分的硫苷,如果作为饲料的主要成分,就可能超过可接受浓度,造成上述的不良反应。[14][15]饲料可以经过处理,洗去硫苷。[16]培育低硫苷菜籽可以提高油渣饼作为饲料的经济价值,是油菜育种的重要方向之一。[17]

十字花科蔬菜在不缺碘的人类无抗甲状腺作用。曾有研究召集志愿者连续四周每天食用150克抱子甘蓝,未造成甲状腺功能异常。在缺碘地区,食用大量十字花科蔬菜可能提高甲状腺癌风险,但在碘摄入正常地区则无此现象。[18]

味觉和饮食行为 编辑

其中,硫代葡萄糖苷黑芥子苷被证明是导致烹熟的花椰菜抱子甘蓝的苦味的原因。[1][19] 硫代葡萄糖苷可能会改变动物的饮食行为。[20]

昆虫 编辑

硫代葡萄糖苷及其产品对许多昆虫具有负面影响,其原因是威慑和毒性的结合。为了在农学背景下应用这一原理,一些硫代葡萄糖苷衍生的产品可以作为拒食剂,即天然杀虫剂[21] 相比之下,十字花科植物的害虫小菜蛾可能会识别出硫代葡萄糖苷的存在,从而可以识别出适当的寄主植物。[22]实际上,在含硫代葡萄糖苷的植物上有一种特有的,专门的昆虫,包括蝴蝶,如欧洲粉蝶菜粉蝶红襟粉蝶,还有某些蚜虫,飞蛾,如南方粘虫,锯蝇和跳蚤甲虫。[来源请求]例如,欧洲粉蝶将卵产在这些含硫代葡萄糖苷的植物上,幼虫即使含有高水平的硫代葡萄糖苷也能存活,并食用含有硫代葡萄糖苷的植物材料。[来源请求]菜粉蝶和都红襟粉蝶具有一种腈特异性蛋白质,它将硫代葡萄糖苷水解转向腈类而不是反应性异硫氰酸酯。[23] 相比之下,小菜蛾具有完全不同的蛋白质,硫代葡萄糖苷硫酸酯酶,它会硫酸化硫代葡萄糖苷,从而使它们不适合黑芥子酶降解为有毒产物。[24]
